I would stick to one florist and either have someone go pick it up the morning of or day before to save the delivery charge. But have them deliver the reception flowers. Most places should include a ~1hr delivery free of charge.
The reasoning for sticking to 1 florist is one less vendor to worry about. And if you order more from 1 florist you may be able to negotiate prices better.

My wedding is in Brooklyn and my reception is in Manhattan - without traffic, we are talking about a 20 minute trip. I have lined up florists I plan to visit after the new year, but am wondering what approach to take.
What I am wondering is if it would cost less to have a Brooklyn florist do the Bklyn flowers (which would include the bouquets, buttonierres, and chuppah) and have a Manhattan florist do the reception site (which would be the centerpieces, cake table, and one or two small pieces for the cocktail area and guestbook table). I mean, if either doesn't have to cross the bridge and go the extra mile, I would think it would be less. Or should I just have one florist do it all?
And I am trying to figure out if I should also add an extra stop, which mean an extra charge, to have the bouquets sent to my place so that I have them for the preceremony pictures, or is this something I can pick up the day before and it'll be okay?
