Why don't you see if there are any anime festivals going on within a 5 hour flight? You could do that as the main surprise, then take him for a combined cooking lesson/fancy dinner? Or if you have more time, do a really nice dinner one night, and then a cooking lesson with a famous chef the next.
Mid-September could be a great time to visit Boston - excellent weather, great restaurants, nice hotels with a range of prices, lots to do - sports, theatre, shows, museums, close to the water (harbor cruises, etc..) and lots more. I wouldn't be surprised if you could find lots of things that would interest him and spoil him for his birthday (what a nice wife!!!)
Good luck coming up with a plan!
Napa is beautiful that time of year! I know you can't enjoy the tastings propery but there are other activities and views to enjoy :)
To keep it local, you could surprise him by renting a theater and invite a bunch of your friends and show some anime flicks and throw in a little cute slideshow. Local theaters would accomodate. Maybe you could cater some food from a favorite restaurant of his.
I'm thinking San Francisco or Napa since you have great food and great wine there. You could fly into SF, go for an amazing dinner, stay at a rad hotel like the Clift and then in the morning have a private car pick you up and take you wine tasting.

Last year when I turned 30, my husband surprised me with an AMAZING birthday extravaganza. He flew me to Vegas, bought me a new cocktail dress and garnet necklace and earrings, took me to see Cirque du Soleil, and took me out to dinner in the Paris restaurant. It was crazy! This year he is turning 30 and I want to make his birthday as special as mine was...except that I don't know how to top that! I need suggestions, ladies!
We live in Chicago and I am pregnant, so I would like to keep any flights to under 5 hours. We will probably travel in mid-September. Husband likes wine, cooking, working out, soccer, dining at restaurants (he knows a lot about all the famous chefs), British television shows, and anime.
Suggestions?
