Are there any inside jokes between you two, or "weird" things that you used to do together when you were growing up, that you could spin into something funny? I would just recommend to keep it clean, and keep it respectful of his new wife and the serious committment that they will have just made.
That being said, I think short, sweet, heartfelt words always win out over looking for a cheap laugh-- I've seen the key thing done before, and it was really just awkward for the "audience" because why would we want to joke about the groom sleeping around with lots of girls on his wedding day, you know?

So, my older brother is getting married two weeks after me. I want to do something cute at the reception like, ha-ha funny to make people laugh and maybe make him a little emb.
Like a good idea I had heard was getting up there, as his siter and saying something along the lines of "So now my big bro is taken off the market, all you ladies gotta turn in the keys to the house, come on, I know your out there, turn in the keys!" and then have like a bunch of random keys given out to girls at the reception to turn in!
I want something kind of like that, something harmless and just funny to mess with him a little, that one wont work, we both are getting married back home and neither one of us live in that state and have not for some years, so it dont really think it will work! Anyone ever seen anything cute or have any good ideas???
