Sugar Bliss on Wabash!! Its in the Loop behind Macy's. They have some great cupcakes. I have to admit that I am a salty snacker and not really into sweets i.e. cakes, cupcakes, cookies but these cupcakes are worth it. Especially the cream cheese frosting! They make cute minis too.

More cupcakes make the best cupcakes in the city, IMHO. Both savory and sweet too, which not all places offer.
I once did a cupcake crawl with some girlfriends, and our two favorites at the end were Sensational Bites on Southport and Molly's Cupcakes on Clark. I'd highly suggest similar "research" excursions with your gals ;)
